Tirzepatide is a synthetic peptide of 39 amino acids engineered from the native glucose-dependent insulinotropic polypeptide sequence. Its structure incorporates several non-natural residues and a C-terminal segment derived from glucagon-like peptide-1, together with a C20 fatty diacid moiety attached through a linker. The lipophilic side chain promotes binding to serum albumin, which slows renal clearance after administration. The compound is classified as a dual incretin receptor agonist and is supplied as a lyophilized powder for reconstitution or as a preformulated solution, depending on the presentation.
The peptide activates two G protein-coupled receptors, GIPR and GLP-1R. Binding triggers adenylyl cyclase activity and raises intracellular cyclic AMP in pancreatic beta cells, which potentiates insulin release when glucose is elevated. Signaling in the central nervous system is associated with reduced appetite and lower energy intake, while effects on gastric emptying and glucagon secretion are also reported. Because activity at both receptors is retained, the pharmacological profile is often described as incretin-based rather than selective for a single receptor.
After subcutaneous injection, absorption is gradual, and peak plasma levels are generally reached within one to three days. Albumin binding extends the apparent half-life to roughly five days, which supports a weekly administration schedule. Metabolism proceeds mainly through proteolytic cleavage of the peptide backbone and beta-oxidation of the fatty acid chain, rather than through cytochrome P450 pathways. Eliminated fragments are largely recycled through general protein turnover, and excretion of intact drug in urine is minimal. These properties distinguish the molecule from short-acting incretin mimetics.
The GIP receptor is expressed in pancreatic islets, adipose tissue, and the central nervous system, while GLP-1 receptors are found in pancreatic islets, the gastrointestinal tract, and the brain. Activation of both receptors can enhance glucose-dependent insulin secretion and reduce glucagon release. The relative contribution of each receptor to the overall pharmacological effect remains an area of ongoing investigation. Preclinical studies suggest that GIP receptor agonism may modulate appetite and energy balance, but the precise mechanisms in humans are not fully established.
In clinical research, tirzepatide has been studied in randomized controlled trials for glycemic control and body weight reduction. These trials typically measure changes in hemoglobin A1c and body weight over periods of several months. The drug is administered by subcutaneous injection, and its pharmacokinetic profile supports once-weekly dosing. Post-marketing surveillance continues to evaluate long-term outcomes and rare adverse events.

Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Development began in the 2010s, when researchers modified a GIP-based scaffold to add GLP-1 activity and then attached the fatty diacid to lengthen its half-life. Clinical evaluation proceeded through large phase 3 programmes in type 2 diabetes and in obesity, and regulators in the United States cleared the compound for type 2 diabetes in 2022 and for chronic weight management in 2023. Several cardiovascular and metabolic outcome studies are still reporting, so the picture of long-term benefit and risk is incomplete. Approvals in other regions followed on different timelines.

Overall, GSK-3 appears to both promote and inhibit apoptosis, and this regulation varies depending on the specific molecular and cellular context. GSK-3 is also involved in nuclear transcriptional activator kappa B (NFκB) signaling pathway, Hedgehog signaling pathway, Notch signaling pathway, and epithelial-mesenchymal transition. Due to its importance across numerous cellular functions, GSK-3 activity is subject to tight regulation and is considered an "Ace" among kinases. The speed and efficacy of GSK-3 phosphorylation is regulated by several factors. Phosphorylation of certain GSK-3 residues can increase or decrease its ability to bind substrate. Phosphorylation at tyrosine-216 in GSK-3β or tyrosine-279 in GSK-3α enhances the enzymatic activity of GSK-3, while phosphorylation of autoinhibitory serine-9 in GSK-3β or serine-21 in GSK-3α significantly decreases active site availability (see figure). Further, GSK-3 is unusual among kinases in that it usually requires a "priming kinase" to first phosphorylate a substrate. A phosphorylated serine or threonine residue located four amino acids C-terminal to the target site of phosphorylation allows the substrate to bind a pocket of positive charge formed by arginine and lysine residues. Depending on the pathway in which it is being utilized, GSK-3 may be further regulated by cellular localization or the formation of protein complexes.
== Antigenic specificity == Antigenic specificity is the ability of the host cells to recognize an antigen specifically as a unique molecular entity and distinguish it from another with exquisite precision. Antigen specificity is due primarily to the side-chain conformations of the antigen. It is measurable and need not be linear or of a rate-limited step or equation. Both T cells and B cells are cellular components of adaptive immunity.
Erasistratus was also responsible for naming and describing the function of the epiglottis and the heart's valves, including the tricuspid. During the third century, Greek physicians were able to differentiate nerves from blood vessels and tendons and to realize that the nerves convey neural impulses. It was Herophilus who made the point that damage to motor nerves induced paralysis. Herophilus named the meninges and ventricles in the brain, appreciated the division between cerebellum and cerebrum and recognized that the brain was the "seat of intellect" and not a "cooling chamber" as propounded by Aristotle. Herophilus is also credited with describing the optic, oculomotor, motor division of the trigeminal, facial, vestibulocochlear and hypoglossal nerves. Incredible feats were made during the third century BCE in both the digestive and reproductive systems. Herophilus discovered and described not only the salivary glands but also the small intestine and liver. He showed that the uterus is a hollow organ and described the ovaries and uterine tubes. He recognized that spermatozoa were produced by the testes and was the first to identify the prostate gland. The anatomy of the muscles and skeleton is described in the Hippocratic Corpus, an Ancient Greek medical work written by unknown authors. Aristotle described vertebrate anatomy based on animal dissection. Praxagoras identified the difference between arteries and veins.
In relation to this, MDMA's duration and the offset of its effects appear to be determined more by rapid acute tolerance rather than by circulating drug concentrations. Similar findings have been made for amphetamine and methamphetamine. One mechanism by which tolerance to MDMA may occur is internalization of the serotonin transporter (SERT). Although MDMA and serotonin are not significant TAAR1 agonists in humans, TAAR1 activation by MDMA may result in SERT internalization, for instance in rodents in whom MDMA is a potent TAAR1 agonist. It is thought that brain serotonin levels are depleted after MDMA administration but that levels typically return to normal within 24 to 48 hours.

Also, in the post-war years, communication between English and German scientists was poor. Following the new reports, the method of gas chromatography spread widely and Cremer's work slowly gained more recognition. Cremer and her students continued their work on developing both the methods and theories behind gas chromatography over the next two decades and led to many of contemporary, common use ideas used in gas chromatography. Cremer and her group created the phrase "relative retention time" and how to calculate the peak area through multiplying the peak's height by the width of the peak at half height. Additionally, they demonstrated the relationship between measurement and column temperature and also invented head space analysis.
The Collaborative for academic, social, emotional learning (CASEL) addresses five broad and interrelated areas of competence and highlights examples for each: self-awareness, self-management, social awareness, relationship skills, and responsible decision-making. A meta-analysis was done by Alexendru Boncu, Iuliana Costeau, & Mihaela Minulescu (2017) looking at social-emotional learning (SEL) studies and the effects on emotional and behavior outcomes. They found a small but significant effect size (across the studies looked into) for externalized problems and social-emotional skills. Holistic approaches to education also emphasize social-emotional development as a key pillar of personal growth.
== Safety == Good clinical compliance was observed at the typical clinical oral dose of 150 mg/day, with no symptomatic side effect reported. The adverse event rate was higher at high dose zinc L-carnosine (300 mg/day) without additional benefits, and therefore high dose is not recommended. Side-effects are associated with the amount of zinc intake. According to the Japanese product monograph, safety in children below the age of 12, pregnant women and lactating women are not established (no experience in use); and the level of use in the elderly population is suggested and recommended at 100 mg zinc L-carnosine per day because of reduced digestive system function in the general elderly population; and those with poor liver functions should be under medical supervision. Those with copper deficiency should also be under medical supervision. Although zinc L-carnosine caused an increase in serum zinc level, the serum copper level and copper:zinc ratio decreased, and a case of preexisting copper deficiency deteriorated. As a mitigative, supplementation of 2 mg/day copper as glycinate chelate safely increases Cu-Zn superoxide dismutase activity. There is no evidence of a reduced tumor response to radiotherapy.

The defeat of the Russian armies forced Vladimir Lenin and the Soviet leadership to postpone their strategic objective of linking up with the German and other European revolutionary leftist collaborators to spread communist revolution. Lenin also hoped for generating support for the Red Army in Poland, which failed to materialize. Piłsudski's seizure of Vilnius in October 1920 (known as Żeligowski's Mutiny) was a nail in the coffin of the already poor Lithuania–Poland relations that had been strained by the Polish–Lithuanian War of 1919–1920; both states would remain hostile to one another for the remainder of the interwar period. Piłsudski's concept of Intermarium (an East European federation of states inspired by the tradition of the multiethnic Polish–Lithuanian Commonwealth that would include a hypothetical multinational successor state to the Grand Duchy of Lithuania) had the fatal flaw of being incompatible with his assumption of Polish domination, which would amount to an encroachment on the neighboring peoples' lands and aspirations. At the time of rising national movements, the plan thus ceased being a feature of Poland's politics.[a] A larger federated structure was also opposed by Dmowski's National Democrats. Their representative at the Peace of Riga talks, Stanisław Grabski, opted for leaving Minsk, Berdychiv, Kamianets-Podilskyi and the surrounding areas on the Soviet side of the border.

White Castle is an American regional hamburger restaurant chain with about 345 locations across 13 states, with its greatest presence in the Midwest and New York metropolitan area. It was founded in 1921, in Wichita, Kansas. White Castle has been generally credited as the world's first fast food hamburger chain. It is known for its small, square hamburgers – commonly referred to as "sliders" – sold at low prices: five cents from their introduction while gradually increasing in price during the 1930s, with promotional coupons in the 1940s, offering five burgers for ten cents. In 2014, Time named the White Castle slider "The Most Influential Burger of All Time". The company's restaurant locations are white buildings decorated with a castle motif.
The establishment of permanent boundaries between what later became Lower Saxony and Westphalia began in the 12th century. In 1260, in a treaty between the Archbishopric of Cologne and the Duchy of Brunswick-Lüneburg the lands claimed by the two territories were separated from each other. The border ran along the Weser to a point north of Nienburg. The northern part of the Weser-Ems region was placed under the rule of Brunswick-Lüneburg. The word Niedersachsen was first used before 1300 in a Dutch rhyming chronicle (Reimchronik). From the 14th century it referred to the Duchy of Saxe-Lauenburg (as opposed to Saxe-Wittenberg). On the creation of the imperial circles in 1500, a Lower Saxon Circle was distinguished from a Lower Rhenish–Westphalian Circle. The latter included the following territories that, in whole or in part, belong today to the state of Lower Saxony: the Bishopric of Osnabrück, the Bishopric of Münster, the County of Bentheim, the County of Hoya, the Principality of East Frisia, the Principality of Verden, the County of Diepholz, the County of Oldenburg, the County of Schaumburg and the County of Spiegelberg. At the same time a distinction was made with the eastern part of the old Saxon lands from the central German principalities later called Upper Saxony for dynastic reasons. The close historical links between the domains of the Lower Saxon Circle now in modern Lower Saxony survived for centuries especially from a dynastic point of view.

Proper nutrition is important after delivery to help the mother recover, and to provide enough food energy and nutrients for a woman to breastfeed her child. Women having serum ferritin less than 70 μg/L may need iron supplements to prevent iron deficiency anaemia during pregnancy and postpartum. During lactation, water intake may need to be increased. Human milk is made of 88% water, and the IOM recommends that breastfeeding women increase their water intake by about 300 mL/day to a total volume of 3000 mL/day (from food and drink), approximately 2,400 mL/day from fluids. It is a synthetic linear peptide that acts as a dual agonist at the GIP and GLP-1 receptors. It combines a modified incretin backbone with a fatty diacid side chain that extends its circulation time. It is not a small-molecule drug and is not orally absorbed in its native form.
Single-receptor agents act predominantly at GLP-1R. Tirzepatide retains activity at GIPR, which is expressed in pancreatic islets, adipose tissue, and the central nervous system. The contribution of each receptor to the overall response is still an area of active study, and the relative weight of GIPR signaling in humans is not fully settled.
The fatty diacid moiety binds serum albumin, which reduces renal filtration of the active peptide. Additional resistance to enzymatic breakdown comes from non-natural residues in the backbone. Together these features produce an apparent half-life of about five days and permit weekly dosing intervals.
It activates both GIP and GLP-1 receptors. This dual action differentiates it from selective GLP-1 agonists.
